Determination of carbofuran in surface water and biological tissue by sol–gel immunoaffinity extraction and on-line preconcentration/HPLC/UV analysis

A selective and simple analytical method for the trace level determination of carbofuran in complex environmental and... more A selective and simple analytical method for the trace level determination of carbofuran in complex environmental and biological samples was developed based on immunoaffinity extraction (IAE) followed by on-line preconcentration and HPLC/UV analysis of the purified extract. The immunosorbent for IAE was prepared by sol–gel encapsulation of monoclonal anti-carbofuran antibodies, and was fully characterized for capacity, repeatability, binding strength, binding kinetics and cross-reactivity. Method performance was evaluated with two different types of difficult samples: dam water and methanolic extracts of epithelial cervical-uterine tissue. Linear behavior and quantitative recoveries were obtained from the analysis of samples spiked with carbofuran at 0.2–4 ng/mL (dam water, 50 mL samples) and 10–40 ng/mL (biological tissue extract, 2 mL samples). RSD (n = 7) and detection limits were, respectively, 10.1% (spike 0.40 ng/mL) and 0.13 ng/mL for dam water; 8.5% (spike 20 ng/mL) and 5 ng/mL for the biological tissue extract. The excellent sample purification achieved with the IAE column allows precise and accurate determination of carbofuran in complex matrices, even when using non-selective UV detection in the chromatographic analysis.

A new macrochromatographic technique was assayed in carboxymethylcellulose and damperer iminazole-HCL-
-KCN-NaCL at 6,7 pH. This technique es reliable and exact for A2 hemoglobin separation and quantization in individuals carrier of AC hemoglobin, C Beta-thalasseamia, AS hemoglobin, and SC hemoglobin, whick, in many cases es of clinical interest. Also, a variant of A2 hemoglobin was detected in a pregnant woman carrier of AC hemoglobin, and a Beta-thalassemic gen in Cis with the gen for C hemoglobin.

Comprehensive two-dimensional gas chromatography (GCxGC) is a major advance for the detailed characterisation of... more Comprehensive two-dimensional gas chromatography (GCxGC) is a major advance for the detailed characterisation of petroleum products. This technique is based on two orthogonal dimensions of separation achieved by two chromatographic capillary columns of different chemistries and selectivities. High-frequency sampling between the two columns is achieved by a modulator, ensuring that the whole sample is transferred and analysed continuously in both separations. Thus, the peak capacity and the resoluting power dramatically increase. Besides, highly structured 2D chromatograms are obtained upon the volatility and the polarity of the solute to provide more accurate molecular identification of hydrocarbons. In this paper fundamental and practical considerations for implementation of GCxGC are reviewed. Selected applications obtained using a prototype of a GCxGC chromatograph developed in-house highlight the potential of the technique for molecular characterisation of middle distillates, sulphur speciation in diesel and analysis of effluents from petrochemical processes.

The detailed characterisation of middle distillates is essential for a better understanding of reactions involved in... more The detailed characterisation of middle distillates is essential for a better understanding of reactions involved in refining process. Owing to higher resolution power and enhanced sensitivity, comprehensive two-dimensional gas chromatography (GC x GC) is a powerful tool for improving characterisation of petroleum samples. The aim of this paper is to compare GC x GC and various ASTM methods - gas chromatography (GC), liquid chromatography (LC) and mass spectrometry (MS) - for group type separation and detailed hydrocarbon analysis. Best features of GC x GC are demonstrated and compared to these techniques in terms of cost, time consumption and accuracy. In particular, a new approach of simulated distillation (SimDis-GC x GC) is proposed: compared to the standard method ASTM D2887 it gives unequal information for better understanding of conversion process.

The effect on sterol stability and CLA content in meats from pigs fed with diets having different contents of oleic... more The effect on sterol stability and CLA content in meats from pigs fed with diets having different contents of oleic and linoleic acid was studied, considering typical Italian products seasoned with or without nitrates and ascorbates. Results showed that the increase of oleic acid in the diet leads to seasoned products with higher contents of oleic acid and lower content of linoleic acid and CLA. A different partition of sterols and sterol oxides was observed in the fat and in the muscle of all the products, with slightly higher amounts of cholesterol and sterol oxides in the muscle. Meat products seasoned in presence of ascorbates showed slightly lower amounts of sterol oxides in respect to those added with nitrates. The statistical treatment of the data showed that fatty acids distribution allow to discriminate between meats from different diets.
